Histopathologic Study on the Antihypertensive Drug Effects On the Renal Arteries of the Spontaneously Hyperteneou.



Abstract
To insist such pathogenic mechanisms, various experimental methods were used. First is Goldblatt¢¥ s kidney, which induce experimental hypertension by narrowing the renal arterial lumen. Second is administration of DOCA, salts and high lipid diet. Third is chemical methods to estimate beta- hydroxybutyrate dehydrogenase, lactic dehydrogenase, non-collagen protein, collagen, protein, glycoprotein etc within the arterial wall. Fourth is morphologic study on the hypertensive artery. Fifth is combination method with administration of therapeutic drugs. Sixth is using spontaneously hypertensive rat (SHR) to study genetic effects on the development of hypertension.
In order to study the pathogenetic mechanism of hypertension, and therapeutic effects the authors selected a combination method. The authors selected SHR (Okamoto-Aoki¢¥s strain) as experimental animal, single and combination administrations of alpha- methyl dopa, Timolol and Labetalol as therapeutic agents, and microscopic examination as observation technique.
Commercially avaiable strains were used as control group for hypertensive experimental group. SHR were devided into II to VlIth group according to the administration of therapuetic agents. The experimental groups were devided as follows.
Group
I Commercially avaiable rat.
II : SHR, daily administrated 200mg/kg. of alphamethyl dopa.
III = SEiR, daily administrated 100mg/kg of alpha - methyl dopa and I mg¢¥¢¥kg of Timolol. N SHR, daily administrated 2 nig/kg of Timolol.
V = SHR, daily administrated 100 mg/kg of alpha-methyl dopa and 25 mg/kg of Labetalol.
VI : SHR, daily administrated 50mg/kg of Labetalol.
VII : SHR, not administrated hypertensive control group.
Blood pressure, heart strok rate and body weight were daily checked and after ~2 weeks dku. ation of experiments, the animal was killed with ether anesthesia.
The kidneys were kept in 10% Formalin for 1 week and then treated as routine histologic technique to stain hematoxylin-eosin double staining, periodic-acid Schiffx¢¥s staining, Massonli trichrome staining and PAS-MT staining.
By the microscopic examination wrinkling of inner elastic membrane of the renal arteriol and vacuolization of the outher muscle layer cells were observed in the treated hypertensive groups. However, its shysiologic meaning could not be clarified by this examination
